What decides the purchase

The compound is usually fixed by the route. These are the things that vary between suppliers and between lots.

  1. Nuclease-free is a test, not an adjective

    The claim means the material has been assayed for DNase and RNase activity and none was detected under stated conditions, with the certificate reporting the method and incubation. 'Molecular biology grade' used loosely sometimes means only high chemical purity. Ask which tests were run — RNase is the one that ruins an RNA experiment and it is remarkably persistent.

  2. Concentration accuracy, because it propagates

    Nucleotide and primer concentrations feed directly into reaction stoichiometry, and an error there shows up as an assay that will not reproduce rather than as an obvious failure. For quantitative work ask how the concentration was determined — spectrophotometrically against a stated extinction coefficient is verifiable, a nominal figure is not.

  3. Freeze-thaw is what actually degrades the lot

    Most reagents in this class are supplied frozen and lose activity through repeated cycling rather than through age. A vial thawed twenty times is not the material on the certificate. Order in unit sizes that let you aliquot once and thaw once, which is usually cheaper than the experiments lost to a degraded stock.

  4. Cold chain has to hold across the whole route

    Dry ice sublimes, and a shipment that spends an unplanned weekend in customs can arrive at ambient temperature with no outward sign. Where the material is temperature-critical, agree the shipping condition and the acceptable transit window at enquiry, and specify whether a temperature indicator should travel with the consignment.

What is the difference between molecular biology grade and nuclease-free?
Molecular biology grade generally denotes high chemical purity suited to the field. Nuclease-free is a specific test result: the material was assayed for DNase and RNase activity and none was detected under the stated conditions. The two often travel together but they are not the same claim, and only the second one tells you an RNA prep is safe.
How much does freeze-thaw cycling actually matter?
Enough to be the usual cause of a reagent that worked last month and does not now. Each cycle costs a little activity, and the loss is cumulative and invisible until an assay drifts. Aliquot on first thaw into single-use volumes, label the date, and treat the aliquot rather than the original vial as the unit of stock.
What happens if a cold-chain shipment is delayed?
Dry ice lasts a defined number of days and then the package warms with no external indication. If a consignment has been held in transit, the safe assumption is that the cold chain broke unless a temperature logger travelled with it and says otherwise. Specifying an indicator at enquiry is inexpensive and turns an unanswerable question into a readable one.
